The essence of the cellular life is the self-reproducing vesicle system coupled with self-replicating information molecules, which is driven by sustainable chemical reaction network. A milestone for creating of the self-reproducing automaton is to develop vesicle growth system coupled with the synthesis of information polymers coded vesicle structure. One candidate for the synthesis of information polymer coding vesicle structure is a template polymerization of polyaniline (PANI) on the surface of dioctyl sulfosuccinate sodium salt (AOT) vesicle, where a emeraldine salt form of PANI (PANI-ES), the half-oxidized state of PANI, is selectively synthesized on the AOT vesicle through a specific interaction between aniline and AOT [1]. In this study, we examine AOT vesicle growth coupled with the template polymerization.
AOT molecules form giant unilamellar vesicles (GUVs) by adjusting the pH in the presence of specific salts. By micro-injecting hydrogen peroxide and AOT micelles toward AOT vesicles in the presence of aniline and horseradish peroxidase isoenzyme C (HRPC), we observed drastic vesicle growth coupled with the synthesis of PANI-ES on the vesicle surface. The PANI-ES polymerization was confirmed by the characteristic VIS/NIR absorption spectral pattern. When we micro-injected only AOT micelles without H2O2, the vesicles showed no growth. These observations indicate that AOT vesicle promotes PANI-ES synthesis as template, and simultaneously, the template polymerization product, PANI-ES, promotes the growth of AOT vesicle. This mutual catalytic relationship between vesicle growth and PANI-ES synthesis will be the first step towards the vesicle based self-reproducing automaton system.
